4. Acrolein Produced from Polyamines Correlated Well with the Diseases Accompanied with Senility

Acrolein, which is highly toxic compound, is produced from polyamines by polyamine oxidase [acetylpolyamine oxidase (AcPAO) and spermine oxidase (SMO)]. We clarified that putrescine, polyamine oxidase activity and protein-conjugated acrolein increased in plasma of patients with renal failure (Fig. 4-1). Moreover, we found good correlation between the values of these markers and the severity of renal failure. The latest study showed that polyamine oxidase activity and protein-conjugated acrolein increased in plasma of patients with stroke (Fig. 4-2). The size of stroke was nearly parallel with the multiplied value of acrolein and polyamine oxidase activity. After the onset of stroke, an increase in AcPAO first occurred, followed by increased level of SMO and finally acrolein. We also succeed to find patients with silent brain infarction (SBI) among normal subjects based on the values of polyamine oxidase activity and protein-conjugated acrolein in plasma. These information would make it possible to do more suitable medical treatments in chronic renal feilure and stroke. Experiments are in progress to find SBI with high probability.
